Progeria An Early Aging Disease

Diseases can arise in many different ways during the pathway of cell replication. A person can also genetically inherit a disease if their parent or past ancestor has the disease. When DNA is replicated it is necessary that it is replicated with accuracy or else mutations will occur. Certain mutations can be neutral while others can be fatal. There are certain proofreading factors and DNA repair mechanisms that correct mutations, but if the mutations are not corrected they can accumulate and possibly cause a deadly disease.

One rare genetic disease that is caused by mutations is progeria, also known as Hutchinson-Gilford Progeria Syndrome, where a person starts to age at an early age. Compared to a normal person, people who have progeria age 8 to 10 times faster. Those affected only live until their teenage years and sometimes to their early 20s.

This disease is possibly yielded by mutations and the DNA damage in somatic cells. Somatic cells are what forms the body of an organism. If the DNA repair mechanism cannot correct these mutations and DNA damage in somatic cells, the mutations and damages will accumulate and lead to aging.

Those who have progeria can actually appear normal at birth but can also start showing the first symptoms at infancy. The symptoms can progressively get worse as the person gets older. These symptoms consist of wrinkled skin, hair loss, kidney failure, loss of eyesight, and cardiovascular problems. Sometimes these patients can have an abnormally large head, protruding eyes, and fragile bones as if they were an old person already. Because patients have a bald scalp it is easy to see the veins in that person's head.

This disease is caused by a point mutation in position 1824 of the LMNA gene. The LMNA gene codes for protein lamin A and protein lamina C which aids in the production of the inner membrane of the cell's nucleus. The mutations causes cytosine to be replaced by thymine, which creates progerin, the shortened form of the prelamin A protein. Progerin then prevents the nuclear lamina from building into the correct structure and therefore hinders the cell from its ability to divide.

A possible treatment is the farnesyl transferase inhibitor which might be able to fix the problem with the progerin preventing the nuclear lamina structure from forming. Although there is no known cure for the disease, there are ways to help those affected with cardiovascular diseases and respiratory problems such as by pass surgery.
